publication of an article in the peer-reviewed Journal of
Neuroinflammation supporting the potential of ASC specks as a
therapeutic target for Parkinson’s disease.
In the paper titled, “ASC specks exacerbate α‑synuclein
pathology via amplifying NLRP3 inflammasome activities,” the
authors reported data on the interplay between ASC speck formation,
NLRP3 inflammasome activation, and pathological progression in a PD
model (induced by human A53T mutant α-synuclein preformed fibrils,
“PFFs”). Following is a summary of key findings:
- There was a direct relationship
between α‑synuclein pathology and ASC expression
- ASC specks enhanced NLRP3
inflammasome activation and reactive microgliosis
- ASC specks accelerated dopaminergic
neuron degeneration and dyskinesia (uncontrolled, involuntary
movement)
- Knockdown of endogenous ASC markedly
suppressed microglial inflammasome activation and neuronal
α‑synuclein aggregation under the challenge of PFFs
The authors stated, “There is a clear association between ASC
specks assembly, NLRP3 inflammasome activation, and PD pathological
progression, especially with regards to α‑synuclein accumulation.

About Inflammasome ASC Inhibitor IC 100
IC 100 is a novel humanized IgG4 monoclonal antibody that
inhibits the inflammasome adaptor protein ASC. IC 100 was designed
to attenuate both initiation and perpetuation of the inflammatory
response. It does so by binding to a specific region of the ASC
component of multiple types of inflammasomes, including NLRP1,
NLRP2, NLRP3, NLRC4, AIM2, Pyrin. Intracellularly, IC 100 binds to
ASC monomers, inhibiting inflammasome formation, thereby blocking
activation of IL-1β early in the inflammatory cascade. IC 100 also
binds to ASC in ASC Specks, both intracellularly and
extracellularly, further blocking activation of IL-1β and the
perpetuation of the inflammatory response that is pathogenic in
inflammatory diseases. Because active cytokines amplify adaptive
immunity through various mechanisms, IC 100, by attenuating
cytokine activation, also attenuates the adaptive immune
response.
